Thereafter, highly electrophilic RMs reacted with nucleophilic groups present in biomolecules such as hepatic proteins, enzymes, and nucleic acids, forming conjugates or adducts, leading to a series of toxicological repercussions. The currently proposed biological pathogenesis, including oxidative stress, mitochondrial damage and dysfunction, endoplasmic reticulum (ER) stress, hepatic metabolic dysregulation, and cellular apoptosis, is depicted. This review concisely updates our knowledge of metabolic activation pathways for seven hepatotoxic EF compounds. Critically, it deepens biochemical understanding of proposed molecular hepatotoxicity mechanisms, providing a theoretical foundation for the strategic use of EF in clinical settings.
